Triple negative and basal-like breast cancer: one or many diseases? Implications for surgical pathologists

چکیده

Take home messages The majority of basal-like breast cancers have a triple negative phenotype and the vast majority of triple negative cancers display a basal-like transcriptome, however the two are not synonymous. Basal-like and triple negative breast cancers comprise a heterogeneous group of diseases, which are characterised by a constellation of morphological features. Not all basal-like and triple negative breast cancers have a poor outcome. The precursors of invasive breast carcinomas of basal-like and triple negative phenotype include ductal carcinoma in situ harbouring a similar phenotype and, possibly, a subgroup of microglandular adenosis. A subgroup of basal-like and triple negative breast cancers displays an exquisite sensitivity to anthracycline-based neoadjuvant chemotherapy. Defects in the p53, pRB and p16 pathways are found in a significant proportion of basal-like and triple negative cancers. BRCA1 pathway dysfunction is found in a substantial proportion of basal-like and triple negative breast cancers and can be exploited therapeutically (e.g. inhibitors of the PARP enzyme and cross-linking agents)

Are triple-negative and basal-like breast cancer synonymous?

To the Editor: In a recent issue of Clinical Cancer Research, Dent and colleagues (1) have reported on the prognostic significance of estrogen negative, progesterone negative, and HER2 negative (triple-negative) class in a large cohort of breast cancer patients. Triple-negative tumors are undeniably one of the most relevant subgroups of breast cancer from a medical oncologist’s perspective, giv...

Biology, metastatic patterns, and treatment of patients with triple-negative breast cancer.

Of the estimated 1 million cases of breast cancer diagnosed annually worldwide, it is estimated that over 170,000 will harbor the triple-negative (estrogen receptor/progesterone receptor/HER2-negative) phenotype. Most, though not all, triple-negative breast cancers will be basal-like on gene expression micorarrays. The basal-like molecular subtype exhibits a unique molecular profile and set of ...

Association of HDAC8 Expression with Pathological Findings in Triple Negative and Non-Triple Negative Breast Cancer: Implications for Diagnosis

Background: Previous data have shown the tumorigenicity roles of HDAC8 in breast cancer. More recently, the oncogenic effects of this molecule have been revealed in TNBC. The present study aimed to determine the diagnostic value of HDAC8 for the differentiation of TNBC from nTNBC tumors. Methods: A total of 50 cancerous and normal adjacent tumor specimens were obtained, and the clinical and pat...

A review of triple-negative breast cancer.

BACKGROUND An estimated 1 million cases of breast cancer are diagnosed annually worldwide. Of these, more than 170,000 are described as triple-negative. Triple-negative breast cancer (TNBC) is defined by the lack of protein expression of estrogen receptor (ER) and progesterone receptor (PR) and the absence of HER2 protein overexpression. TNBC is a subtype of breast cancer that overlaps with the...

Tumor cell-produced matrix metalloproteinase 9 (MMP-9) drives malignant progression and metastasis of basal-like triple negative breast cancer

Matrix metalloproteinases (MMPs) have been implicated in diverse roles in breast cancer development and progression. While many of the different MMPs expressed in breast cancer are produced by stromal cells MMP-9 is produced mainly by the tumor cells themselves. To date, the functional role of tumor cell-produced MMP-9 has remained unclear. Here, we show that human breast cancer cell-produced M...
